Fold-change in KD for miRNA puzzles The fold‐change in KD for a switch is the ratio of the KD in the OFF ‐state over the KD in the ON‐state. A  good switch has a lower KD in the ON‐state, so a fold‐change greater than 1 indicates a design that  switches in the right direction. In the figures below, we find that, on average, the “Sensor v2, turn‐off  variant” puzzles exhibit a fold‐change lower than 1. This means that most designs actually switch in the  wrong direction. The “good” switches are found in the long tails to the right. For “Sensor for hsa‐mir‐ 208a”, this tail seems more pronounced with some switches almost reaching a 100‐fold change in KD.  Sensor for hsa-mir-208a
